Nó bao gồm việc cho mẫu chứa chất cần phân tích trong "pha động", thường là dòng chảy của dung môi, di chuyển qua "pha tĩnh." Pha tĩnh trì hoãn sự di chuyển của các thành phần trong mẫu. Khi các thành phần này di chuyển qua hệ thống với tốc độ khác nhau, chúng sẽ được tách khỏi nhau theo thời gian, giống như các vận động viên chạy maratông. Một cách lí tưởng, mỗi thành phần đi qua hệ thống trong một khoảng thời gian riêng biệt, gọi là "thời gian lưu." Trong kĩ thuật sắc ký, hỗn hợp được chuyên chở trong chất lỏng hoặc khí và các thành phần của nó được tách ra do sự phân bố khác nhau của các chất tan khi chúng chảy qua pha tĩnh rắn hay lỏng. Nhiều kĩ thuật khác nhau đã được dùng để phân tích hợp chất phức tạp dựa trên ái tính khác nhau của các chất trong môi trường động khí hoặc lỏng và đối với môi trường hấp phụ tĩnh mà chúng di chuyển qua, như giấy, gelatin hay gel magnesium silicate. Sắc ký phân tích được dùng để xác định danh tính và nồng độ các phân tử trong hỗn hợp. Sắc ký tinh chế được dùng để tinh chế các chất có trong hỗn hợp. Sơ lược về hệ thống GC Mẫu được bơm vào trong và theo dòng khí mang (khí mang thường là N2) đưa đến cột sắc ký (pha tĩnh). Mẫu khi qua cột này sẽ được hấp phụ lên trên pha tĩnh đó. Sau đó, các chất lần lượt tách khỏi cột theo dòng khí ra ngoài được ghi nhận bởi đầu dò. Từ các tín hiệu nhận được máy tính sẽ xử lý và biểu hiện kết quả bằng sắc ký đồ. Nguồn cung cấp khí mang: Có thể sử dụng bình chứa khí hoặc các thiết bị sinh khí (thiết bị tách khí N2 từ không khí, thiết bị cung cấp khí H2 từ nước cất,…). 1.2. Lò cột: dùng để điều khiển nhiệt độ cột phân tích 1.3. Bộ phận tiêm mẫu Bộ phận tiêm mẫu dùng để đưa mẫu vào cột phân tích theo với thể tích bơm có thể thay đổi. Khi đưa mẫu vào cột, có thể sử dụng chế độ chia dòng (split) và không chia dòng (splitless). Có 2 cách đưa mẫu vào cột: bằng tiêm mẫu thủ công và tiêm mẫu tự động (Autosamper – có hoặc không có bộ phận hóa hơi - headspace). 1.4. Cột phân tích Có 2 loại cột: cột nhồi và cột mao quản. - Cột nhồi (packed column): pha tĩnh được nhồi vào trong cột, cột có đường kính 2-4mm và chiều dài 2-3m. - Cột mao quản (capillary): pha tĩnh được phủ mặt trong (bề dày 0.2-0.5µm), cột có đường kính trong 0.1-0.5mm và chiều dài 30-100m. Có nhiều loại đầu dò khác nhau tùy theo mục đích phân tích như đầu dò ion hóa ngọn lửa (FID-Flame Ioniation Detetor), đầu dò dẫn nhiệt (TCD-Thermal Conductivity Detector), đầu dò cộng kết điện tử (ECD-Electron Capture Detector), đầu dò quang hóa ngọn lửa (FPD-Flame Photometric Detector), đầu dò NPD (NPD-Nitrogen Phospho Detector), đầu dò khối phổ (MS-Mass Spectrometry). 1.6. Bộ phận ghi nhận tín hiệu Bộ phận này ghi tín hiệu do đầu dò phát hiện. Đối với các hệ thống HPLC hiện đại, phần này được phần mềm trong hệ thống ghi nhận, lưu các thông số, sắc ký đồ, các thông số liên quan đến peak như tính đối xứng, hệ số phân giải,… đồng thời tính toán, xử lý các thông số liên quan đến kết quả phân tích. 1.7. In dữ liệu Sau khi phân tích xong, dữ liệu sẽ được in ra qua máy in kết nối với máy tính có cài phần mềm điều khiển.
